Introducción: En la electroforesis desnaturalizante en gel de Poliacrilamida son escasos los estudios donde se compare el efecto del tamaño del gel en la separación electroforética, manteniendo la composición química y condiciones de corrida. Objetivos: Evaluar si la cámara de menor formato mini-PROTEAN Tetra cell puede sustituir al modelo PROTEAN II sin pérdida de calidad analítica, reduciendo así el tiempo y costo del análisis de pureza en control de la calidad. Materiales y Métodos: Se evaluó la resolución de las bandas y la pureza de 7 lotes de Interferón Gamma humano recombinante. Se compararon parámetros como precisión, linealidad y límite de detección. Resultados: Se observó comparable resolución de las bandas, y el análisis de ANOVA no mostró diferencias significativas entre los resultados de pureza obtenidos. El parámetro de precisión no indicó una variabilidad significativa entre dimensiones y se obtuvo un coeficiente de variación menor al 1% entre las réplicas para cada gel. En la linealidad se obtienen valores de regresión similares y superiores a 0,98. El límite de detección demostró que ambos geles poseen igual sensibilidad con un límite de 0.1 µg. Conclusiones: Los resultados para ambas dimensiones, demuestran que no existen diferencias significativas entre ambos formatos de cámaras. Los parámetros de validación evaluados indicaron que no existe impacto negativo con el cambio, demostrando la posibilidad de reemplazar el formato PROTEAN II por el formato mini-PROTEAN Tetra cell obteniéndose con esto una disminución de tiempo de ensayo y de gastos involucrados dado por el menor consumo de soluciones y reactivos.

Abstract
Introduction: In denaturing polyacrylamide gel electrophoresis, there are few studies comparing the effect of gel size on electrophoretic separation while maintaining chemical composition and running conditions. Objectives: To evaluate whether the smaller mini-PROTEAN Tetra cell chamber can replace the PROTEAN II model without loss of analytical quality, thus reducing the time and cost of purity analysis in quality control. Materials and Methods: The band resolution and purity of seven batches of recombinant human interferon gamma were evaluated. Parameters such as precision, linearity, and limit of detection were compared. Results: Comparable band resolution was observed, and ANOVA analysis showed no significant differences between the purity results obtained. The precision parameter did not indicate significant variability between dimensions, and a coefficient of variation of less than 1% was obtained between replicates for each gel. Linearity showed similar regression values, exceeding 0.98. The detection limit showed that both gels have equal sensitivity, with a limit of 0.1 µg. Conclusions: The results for both dimensions demonstrate that there are no significant differences between the two chamber formats. The evaluated validation parameters indicated no negative impact from the change, demonstrating the feasibility of replacing the PROTEAN II format with the mini-PROTEAN Tetra cell format, resulting in reduced testing time and expenses involved due to lower consumption of solutions and reagents.
